She is most known for her character work and celebrity impressions on the sketch comedy shows Saturday Night Live (2012–2022) and The Big Gay Sketch Show (2007–2010), where she was a cast member for ten seasons.

Balls Out (2014), Ghostbusters (2016), Office Christmas Party (2016), Rough Night (2017), The Spy Who Dumped Me (2018), Yesterday (2019), and Bombshell (2019) are some of her cinematic credits (2019).

In the miniseries Joe vs. Carole, she portrays Tiger King subject Carole Baskin in 2022.

McKinnon has been nominated for nine Primetime Emmy Awards, winning in 2016 and 2017 for Outstanding Original Music and Lyrics and Outstanding Supporting Actress in a Comedy Series.
